Abstract
目的 探讨雷帕霉素(rapamycin,RAPA)诱导的自噬对人牙髓干细胞(human dental pulp stem cells,hDPSCs)体外增殖的影响.方法 以不同浓度(1 nM,2 nM,5 nM,10 nM,20 nM)RAPA处理hDPSCs,GFP-LC3融合蛋白示踪实验及蛋白免疫印迹法检测hDPSCs自噬活性及自噬相关蛋白表达水平;CCK-8法检测细胞的增殖率变化;流式细胞仪检测细胞周期变化;蛋白免疫印迹法检测细胞增殖及细胞周期相关蛋白表达水平.结果 不同浓度RAPA均可诱导hDPSCs自噬.与对照组相比,诱导组细胞内自噬小体数量显著增多,自噬相关基因5(ATG5)、P62、脂质化轻链蛋白3(LC3-Ⅱ/Ⅰ)表达显著增高(P<0.001).自噬可显著抑制细胞增殖(P<0.001),改变细胞周期时相,G0/G1期细胞比例升高(正常63.5%,实验组83.4%~89.9%),细胞增殖指数显著降低(正常36.5%,实验组16.6%~10.1%),作用与药物浓度呈正相关.与对照组相比,实验组细胞周期蛋白D1(Cyclin D1)、周期素激酶4(CDK4)、周期素激酶6(CDK6)及细胞增殖核抗原(proliferating cell nuclear antigen,PCNA)的表达显著下调(P<0.001).结论 RAPA诱导的细胞自噬可显著抑制hDPSCs的体外增殖,其作用与细胞周期改变和细胞增殖相关蛋白表达下调有关.
