Abstract | 目的 探讨恩格列净(EMPA)在心肌缺血再灌流(MI/R)损伤中的作用及机制.方法 利用大鼠离体心脏缺血40 min再灌流2 h模型,40只雄性SD大鼠随机分为:对照组(Control)、MI/R组、2.5 mol/L EMPA+MI/R组(EMPA+MI/R)以及EMPA+10 mol/L沉默信息调节因子1(SIRT1)抑制剂EX527+MI/R组(EMPA+EX527+MI/R),记录心脏功能、心肌纤维形态、心肌梗死面积,ELISA检测心肌中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和琥珀酸脱氢酶(SDH)活性,Western blot检测Cytochrome c、Cleaved caspase-3、SOD2、gp91 phox和SIRT1蛋白表达,DHE染色检测ROS的含量.结果 和对照组比较,MI/R组心脏功能降低,心肌纤维断裂,心肌梗死面积、Cytochrome c和Cleaved caspase-3以及gp91 phox表达、MDA含量均增加,而SOD和SDH活性以及SOD2和SIRT1表达减少,ROS含量增加;和MI/R组比较,EMPA+MI/R组能提高心脏功能,减轻心肌纤维断裂,降低心肌梗死面积、Cytochrome c、Cleaved caspase-3和gp91 phox表达以及MDA含量,并能增加SOD和SDH活性以及SOD2和SIRT1表达,同时降低ROS含量;然而,EX527可以逆转EMPA对MI/R心脏的保护作用.结论 EMPA激活SIRT1抑制线粒体氧化应激和细胞凋亡,减轻MI/R损伤. |
