Abstract
本发明提供了一种快速制备高通量测序文库的方法及应用。本发明提供的技术方案基于我们对该技术流程的深入研究,对加adapter的反应体系进行了优化,提高了加adapter的效率,降低了文库构建成本及人为操作误差出现的可能性。此外,我们还选择了最优的PCR反应体系,利用有些酶(如Taq酶)加A的特性,达到模版扩增的同时对PCR产物末端加A的效果,相比当前将模板扩增与加A步骤分开进行的方法,本发明大大的减少了整个构库时间。
Other Abstract
The invention provides a method for rapid preparation of a high-throughput sequencing library and application. The technical scheme provided by the invention is based on the in-depth study of the technical process, optimizes an adapter added reaction system, improves the adapter adding efficiency, and reduces the library construction cost and possibility of artificial operation error. In addition, the invention selects an optimal PCR reaction system, and utilizes the A adding characteristic of some enzymes (like Taq enzyme) to reach the effects of template amplification and adding of A to a PCR product end simultaneously. Compared with the current method of conducting template amplification and A adding separately, the method provided by the invention greatly reduces the whole library construction time.
