本发明提供一种检测古菌细胞膜脂中带完整极性头化合物的方法,包括以下步骤:在MRM下将色谱柱温度设定为40~50℃,流速为0.15~0.25mL/min;控制离子源干燥气流速2.5~3.5L/min,温度280~320℃;鞘气流速8.5~9.5L/min,温度280~320℃;喷雾压力400~450kPa;毛细管电压3.5~5.0kV;喷射电压650~750V;碎裂反应的电压150~250仪器单位,碰撞能量10~50仪器单位;不带任何极性头的核心脂部分质荷比为1302.3~1292.3;进行古菌细胞膜脂中带完整极性头化合物的检测。本发明具有检测灵敏度高、检出限低的特点。
Other Abstract
The invention provides a method of detecting a compound with an intact polar head in archaea cell membrane lipid. The method comprises the steps that under MRM, chromatographic column temperature is set to be 40-50 DEG C, and the flow velocity is set to be 0.15-0.25mL/min; the flow velocity of an ion source drying gas is controlled to be 2.5-3.5L/min, and the temperature of the ion source drying gas is controlled to be 280-320 DEG C; the flow velocity of a sheath gas is controlled to be 8.5-9.5L/min, and the temperature of the sheath gas is controlled to be 280-320 DEG C; spray pressure is controlled to be 400-450kPa; the capillary voltage is controlled to be 3.5-5.0kV; the jet voltage is controlled to be 650-750V; the voltage of a fragmentation reaction is controlled to be 150-250 instrument units, and collision energy is controlled to be 10-50 instrument units; the mass-to-charge ratio of a core lipid part without any polar head is 1,302.3-1,292.3; and detection of the compound withthe intact polar head in the archaea cell membrane lipid is performed. The method has the advantages of being high in detection sensitivity and low in detection limit.
