Abstract | 本发明提供一种探针及其制备方法和应用。该探针包括核酸片段,核酸片段通过多臂修饰性PEG标记有荧光物质。核苷酸通过多臂修饰性PEG实现PEG化,多臂修饰性PEG的修饰基团与荧光物质发生偶联反应从而使荧光物质标记到核酸片段上。以此形成的探针中,单个核苷酸有多个修饰基团形成的偶联位点,更多的偶联位点可以结合更多的荧光物质。因而,标记后的探针其单位长度上的荧光物质更多,检测时表现出的荧光强度更高,从而能够有效提高检测的灵敏度和精确性。 |
Other Abstract | The invention provides a probe, and a preparation method and application thereof. The probe comprises a nucleic acid fragment, and the nucleic acid fragment is labeled with a fluorescent substance through multi-arm modified PEG. Nucleotide realizes PEGylation through the multi-arm modified PEG, and a modification group of the multi-arm modified PEG and the fluorescent substance undergo a couplingreaction so as to allow the fluorescent substance to be labeled on the nucleic acid fragment. In the probe formed in the invention, a single nucleotide has coupling sites formed by a plurality of modification groups, and more coupling sites can be combined with more fluorescent substances. Thus, the labeled probe has more fluorescent substances in unit length and shows higher fluorescence intensity during detection, so detection sensitivity and accuracy can be effectively improved. |
